چکیده

The adsorption and inhibitory action of pyocin 21 on sensitive cells of Pseudonzonas aeruginosa (P. pyocyanea) were observed by fluorescence microscopy and by using agar cultures. By electron microscopy, preparations from four pyocinogenic strains of P. aeruginosa were seen to include contracted and uncontracted particles, isolated tail cores and sheaths, The importance of adsorption and contraction of the particles on the surface of sensitive bacteria for the inhibitory action of these pyocins is suggested by the following evidence : ( I ) Particles adsorbed to sensitive bacteria a t o "C, but contraction of particles was not then observed and the bacteria were not disrupted. ( 2 ) Pyocin particles did not adsorb to pyocin-resistant bacteria. (3) The ability to adsorb to sensitive bacteria and to kill them was absent after removal of the contractile sheath with sodium dodecyl sulphate. (4) When treated with homologous antiserum, pyocin activity was neutralized and the agglutinated particles remained unadsorbed and uncontracted. ( 5 ) Bacterial receptors for contractile pyocins contain lipopolysaccharide ; such extracts from sensitive, but not from resistant, cells of P. aeruginosa inactivated pyocins, and contracted particles were seen adsorbed to lipopolysaccharide fragments.

Atomic Structure of Type VI Contractile Sheath from Pseudomonas aeruginosa

Pseudomonas aeruginosa has three type VI secretion systems (T6SSs), H1-, H2-, and H3-T6SS, each belonging to a distinct group. The two T6SS components, TssB/VipA and TssC/VipB, assemble to form tubules that conserve structural/functional homology with tail sheaths of contractile bacteriophages and pyocins. Here, we used cryoelectron microscopy to solve the structure of the H1-T6SS P. aeruginosa...

Effect of environment on sensitivity of Neisseria gonorrhoeae to Pseudomonas aeruginosa bacteriocins.

The effect of environmental variation on the susceptibility of Neisseria gonorrhoeae to pyocin produced by Pseudomonas aeruginosa was examined. Susceptibility to at least one pyocin was demonstrated in strains of N. gonorrhoeae (99%), N. meningitidis (35%), and N. lactamica (47%). The degree of sensitivity to pyocin displayed by N. gonorrhoeae was affected by varying the pH of the growth enviro...

Antibacterial efficacy of R-type pyocins towards Pseudomonas aeruginosa in a murine peritonitis model.

R-type pyocins are high-molecular-weight bacteriocins carried within the chromosomes of some bacterial species, such as Pseudomonas aeruginosa, and almost certainly evolved from lysogenic bacteriophages of the Myoviridae family. They contain no head structures and no DNA and are used as defense systems, usually against other strains of the same bacterial species. They bind with their tail fiber...

Lipopolysaccharide as shield and receptor for R-pyocin-mediated killing in Pseudomonas aeruginosa.

Pseudomonas aeruginosa produces three different types of bacteriocins: the soluble S-pyocins and the bacteriophage-like F- and R-pyocins. R-pyocins kill susceptible bacteria of the same or closely related species with high efficiency. Five different types of R-pyocins (R1- to R5-pyocins) have been described based on their killing spectra and tail fiber protein sequences. We analyzed the distrib...

Plant-expressed pyocins for control of Pseudomonas aeruginosa

The emergence, persistence and spread of antibiotic-resistant human pathogenic bacteria heralds a growing global health crisis. Drug-resistant strains of gram-negative bacteria, such as Pseudomonas aeruginosa, are especially dangerous and the medical and economic burden they impose underscore the critical need for finding new antimicrobials. Recent studies have demonstrated that plant-expressed...
